Immunocytochemistry analysis of HeLa cells labeling c-Fos with ab190289 at 1/5000 dilution (red) and mouse mAb to tubulin at 1/10000 dilution (green). The blue is DAPI staining of nuclear DNA. HeLa cells were kept in fetal bovine serum (FBS) – free media for 36 hours. Then the cells were treated with PBS (A) as a control or stimulated with 20% FBS (B) for 30 min. c-FOS antibody labels only the nuclei of stimulated cells.

Following transcardial perfusion of mouse with 4% paraformaldehyde, brain was post fixed for 24 hours, cut to 45μM, and free-floating sections were stained with above antibodies. The c-FOS antibody stains only nuclei of spontaneously active neurons. NF-L is expressed in axons of neuronal cells.

Pregnenolone (Preg) inhibited the RANKL-induced activation of ERK and NF-κB signaling cascades, and the downstream induction of c-Fos and NFATc1. (A) To examine early RANKL signaling events, BMMs were serum-starved for 1 h, pre-treated with 20 μM Preg for 3 h, and then stimulated with RANKL for the indicated time. Total cellular protein were extracted and subjected to western blot analyses using specific antibodies against ERK and p-ERK, p38 and p-p38, JNK and p-JNK, IκBα, NF-κB p65 and p-p65, and β-actin. (B–F) Quantitative densitometric analysis of phosphorylated protein to total protein counterpart, or normalized to β-actin were conducted. (G) To examine downstream (late stage) RANLK signaling events, BMMs were treated with RANKL for 0, 1, 3, and 5 d. M-CSF-dependent BMMs were stimulated with RANKL without or with 10 and 20 μM Preg for 72 h. Total cellular proteins were extracted and subjected to western blot analyses using specific antibodies against NFATc1, c-Fos, and GAPDH.
