Immunocytochemistry/ Immunofluorescence - Anti-AP2 alpha + beta antibody [A6/2/2] (AB61)

ICC/IF image of ab61 stained HeLa cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal Goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab61, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 Goat anti-Mouse IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

AP2 alpha + beta also known as adaptor protein complex 2 functions as part of the protein coat complex that facilitates clathrin-mediated endocytosis. Composed of alpha and beta subunits this complex has an approximate mass of 200 kDa. You can find AP2 alpha + beta highly expressed in cells with active endocytic processes such as neurons and epithelial cells.
Biological function summary

AP2 alpha + beta mediates the recruitment and assembly of clathrin-coated vesicles at the plasma membrane. This complex interacts directly with clathrin and cargo receptors playing an important role in cargo selection during endocytosis. It has an important role in cellular nutrient uptake and receptor internalization which are necessary for maintaining cellular function and signaling.

Pathways

Functions of AP2 alpha + beta are integral to the endocytic pathway and intracellular trafficking. It interacts with proteins such as clathrin and dynamin to ensure proper vesicle maturation and scission. You can find it involved in pathways that regulate cell surface receptor levels and synaptic vesicle recycling highlighting its interaction with proteins like AP180 and Eps15.

AP2 alpha + beta involvement correlates with neurological disorders and cancer. Abnormal function or expression of these proteins has been observed in Alzheimer’s disease where altered endocytosis affects amyloid precursor protein processing. Additionally AP2 alpha + beta mutations or dysregulation has connections to tumor progression often linked with oncogenic receptor endocytosis.

Sequence-specific DNA-binding protein that interacts with inducible viral and cellular enhancer elements to regulate transcription of selected genes. AP-2 factors bind to the consensus sequence 5'-GCCNNNGGC-3' and activate genes involved in a large spectrum of important biological functions including proper eye, face, body wall, limb and neural tube development. They also suppress a number of genes including MCAM/MUC18, C/EBP alpha and MYC. AP-2-alpha is the only AP-2 protein required for early morphogenesis of the lens vesicle. Together with the CITED2 coactivator, stimulates the PITX2 P1 promoter transcription activation. Associates with chromatin to the PITX2 P1 promoter region.
