      Coatomer proteins are involved in regulating transport between the endoplasmic reticulum (ER) and the Golgi complex and in intra-Golgi transport. There exist two coatomer-protein mechanisms (COP I and COP II) and although they have mechanistic parallels, they are molecularly distinct. The COP I coat is comprised of seven subunits (alpha-, beta-, beta'-, gamma-, delta-, epsilon-, and zeta-COP) in a complex called coatomer. Assembly of the coatomer (COP I) onto non-clathrin coated vesicles is regulated by ADP-ribosylation factor (ARF). Vesicle formation, budding, fusion, and disassembly is dependent on GDP-GTP exchange, COP I, and ARF. COP I has been shown to facilitate retrograde intracellular transport from the ER to the Golgi complex. By contrast, COPII facilitates anterograde transport between these subcellular organelles. COP II has been shown to be independently and selectively recruited to the ER relative to COP I subunits.

      The coatomer is a cytosolic protein complex that binds to dilysine motifs and reversibly associates with Golgi non-clathrin-coated vesicles, which further mediate biosynthetic protein transport from the ER, via the Golgi up to the trans Golgi network. Coatomer complex is required for budding from Golgi membranes, and is essential for the retrograde Golgi-to-ER transport of dilysine-tagged proteins. In mammals, the coatomer can only be recruited by membranes associated to ADP-ribosylation factors (ARFs), which are small GTP-binding proteins; the complex also influences the Golgi structural integrity, as well as the processing, activity, and endocytic recycling of LDL receptors.
      Xenin stimulates exocrine pancreatic secretion. It inhibits pentagastrin-stimulated secretion of acid, to induce exocrine pancreatic secretion and to affect small and large intestinal motility. In the gut, xenin interacts with the neurotensin receptor.

    Here's the detailed explanation: we bought 2 antibody from you company , alpha-COP and delta-COP, these two protein are both subunits of COPI vesicle and they both in out layer of COPI , so it means these two proteins should localized at pretty similar position in cells, both outside nuclei, golgi-like morphology. we used these two antibodies for confocal experiment on Hela cells, we got quite different result for these two antibody, the delta-COP one work well, which has right morphology while the alpha-COP showed wield localization in the cell, which is inside nuclei. I attached the confocal data (Image) and protocol . based on the known knowledge, there is something wrong with the alpha-COP antibody, please help us to solve this problem.     Thank you for the explanation, and for the image and protocol. The staining you are seeing with the alpha COP anditboy ab2913 does look incorrect, if all the staining is expected to be peri-nuclear, as is seen with the delta COP antibody. The small black and white image of HeLa cells on the ab2913 datasheet also shows what appears to be intra-nuclear staining, along with the ER/Golgi stain. This suggests that the antibody routinely cross-reacts with a nuclear protein that is not alpha COP (unless the apparent nuclear stain is actually COP on top of the nuclei). We have not received other complaints, but few people have tried ab2913. I suggest replacing the antibody with another, but the only other alpha COP antibody in our catalogue is ab104710, and this has only been tested for western blotting. It may work for ICC, but you will need to optimize the stain. If you are interested in trying this, I will send a vial free-of-charge, to replace ab2913.     I am interested in your product,alpha COP antibody (ab2913, and I have question about this. Do you have any Western Blotting data tested to the antibody? What I want to know is that the antibody detect unknown protein, or the alpha cop antibody detect only for alpha COP. I read the datasheet on the web-site. I learn your product is a polyclonal antibody. If the antibody could detect,which size potein and how many kinds of unknowm (or non-specific)the antibody detect on the Western Bottling.     Thank you for your email. Ab2913 was tested for application in Western blotting using PC-12 cell extract. The antibody detected a band at approximately 140 kDa representing COPI alpha. The antibody also detected a band at approximately 100 kDa, the identity of which is unknown. We currently do not have an image, but we do guarantee that the antibody will work in Western blotting and cross-react with human and rat alpha COP I. I would recommend using PC-12 cell extract as a positive control and we also sell the blocking peptide (ab4931) which can be used in control experiments.     I'm so sorry but I miswrote the application what our customer used. I wrote that they demonstrated the 'Immunocytochemistry' using #AB2913, Indeed, they demonstrated the 'Immunocytometry' by using this antibody, not 'Immunohistchemistry' I describe the detail of their experimental condition as follows. They demonstrated 'Immunocytometry' using #AB2913. The Lot# was 51911 In their experiment, they could not detect any fluorescence signal. They stained the HeLa cell and did not fix the samples. The ratio of the dilution of primary antibody was 1:300. Reaction was performed at 37 degree centigrade for 90 minutes. The anti-goat IgG antibody (raised in Rabbit) which conjugated with Alexa Fluor 488 (Molecular Probe Inc.) was used for the secondary antibody. The ratio of the dilution of secondary antibody was 1:100. They already checked the secondary antibody worked well using anti- AP-2 and beta-COP antibodies as the primary antibody . Positive and negative control was not used.
