While we have validated ab9399 and ab7634 for use in ELISA, this has been done in indirect and direct ELISA respectively, where an antigen was coated to the plate and these antibodies were used separately for detection. These antibodies have not been validated for use in a sandwich ELISA (sELISA) format.

Thank you for your inquiry. The alpha 1 Antitrypsin (AAT) antibody ab9399 is raised against the full-length human protein. However, the epitope has not been mapped and it is therefore not possible to identify the sequence which is recognised by this mouse monoclonal antibody. Indeed, I can confirm that the sequence similarity between the human and hamster AAT is 70%. Unfortunately, given this low homology we would not expect this antibody to cross-react with hamster, and hence we would not recommend using ab9399 as a control with this species. All of our antibodies are tested and guaranteed to work by direct ELISA against the antigen that they were raised. However, searching through the antibodies in our catalogue I can tell you that we stock an alpha 1 antitrypsin antibody (ab8768) that has received a fantastic Abreview by one of our customers for its application by ELISA. I would like to recommend that this antibody is used as the detection antibody (HRP conjugated) and that you employ a monoclonal antibody such as ab9400 alpha 1 Antitrypsin antibody [G11] for the capture of the antigen.
