Flow Cytometry (Intracellular) - Alexa Fluor® 647 Anti-Myc tag antibody [9E10] (AB223895)
Flow cytometry analysis of HEK-293T (human embryonic kidney) transfected with Myc-tagged TMEM119 expression vector labelling Myc-tag with ab223895 at 1/500 dilution (right) compared with Mouse IgG1 (monoclonal) Alexa Fluor® 647 (left). Cells were surface-stained with TMEM119 antibody (ab210405), then fixed with 2% PFA for 10 minutes and permeabilised with 0.1% Tween-20 for 30 minutes. Next, they were stained with Alexa Fluor® 647 conjugated Myc-tag antibody (ab223895) and Alexa Fluor® 488 conjugated anti-Rabbit secondary antibody (for TMEM119). Only TMEM119 (+) population showed Myc-tag positive staining.

ab223895 staining Myc-tag (pseudocolored in white) in 293T cells transfected with TMEM119 with myc tag. The cells were fixed with 4% formaldehyde (10 min), permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab223895 at 1/100 dilution and ab195889, Mouse monoclonal to alpha Tubulin (Alexa Fluor® 594), at 1/250 dilution (shown in red). Nuclear DNA was labelled with DAPI (shown in blue) and Alexa Fluor® 488 conjugated control Myc-tag antibody (different clone) is shown in green.
Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).
