Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Alexa Fluor® 647 Anti-IL-12RB1 antibody [EPR23693-37] (AB282729)

Immunohistochemical analysis of paraffin-embedded human tonsil tissue labelling IL-12RB1 with ab282729 at 1/100 dilution (5.0 µg/ml). Positive staining on human tonsil. The section was then incubated overnight at +4°C in TBS containing 0.025% (v/v) Triton X-100 and 1% (w/v) BSA with ab282729 at 1/100 dilution (shown in red). Nuclear DNA was labeled with DAPI (shown in blue). The section was then mounted using Fluoromount®. Non-specific protein-protein interactions were then blocked in TBS containing 0.025% (v/v) Triton X-100, 0.3M (w/v) glycine and 1% (w/v) BSA for 1h at room temperature. Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8). Counterstained with DAPI.

Immunohistochemical analysis of paraffin-embedded human skeletal muscle tissue labelling IL-12RB1 with ab282729 at 1/100 dilution (5.0 µg/ml). Negative control : no staining on human skeletal muscle. The section was then incubated overnight at +4°C in TBS containing 0.025% (v/v) Triton X-100 and 1% (w/v) BSA with ab282729 at 1/100 dilution (shown in red). Nuclear DNA was labeled with DAPI (shown in blue). The section was then mounted using Fluoromount®. Heat mediated antigen retrieval using sodium citrate buffer (10mM citrate pH 6.0 + 0.05% Tween-20).

靶点信息

Functions as an interleukin receptor which binds interleukin-12 with low affinity and is involved in IL12 transduction. Associated with IL12RB2 it forms a functional, high affinity receptor for IL12. Also associates with IL23R to form the interleukin-23 receptor which functions in IL23 signal transduction probably through activation of the Jak-Stat signaling cascade.
