The aryl hydrocarbon receptor repressor (AHRR) is a protein that functions by inhibiting the activity of the aryl hydrocarbon receptor (AHR). It achieves this by competing with AHR for binding to AHR nuclear translocator (ARNT) forming a complex that prevents AHR from activating its target genes. AHRR weighs approximately 78 kDa and is expressed in various tissues particularly in those with a high exposure to environmental toxins such as the liver and lungs. This expression pattern suggests its role in modulating responses to external chemicals.
Biological function summary

AHRR is essential in regulating gene expression involved in responses to xenobiotics. It is part of the AHR signaling complex functioning as a feedback inhibitor that controls the extent and duration of AHR-mediated transcriptional activity. Mediates dioxin toxicity and is involved in regulation of cell growth and differentiation. Represses the transcription activity of AHR by competing with this transcription factor for heterodimer formation with the ARNT and subsequently binding to the xenobiotic response element (XRE) sequence present in the promoter regulatory region of variety of genes. Represses CYP1A1 by binding the XRE sequence and recruiting ANKRA2, HDAC4 and/or HDAC5. Autoregulates its expression by associating with its own XRE site.
