IHC image of Adenine Nucleotide Translocase staining in Human normal heart muscle formalin fixed paraffin embedded tissue section, performed on a Leica Bond™ system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab109864, 5μg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Adenine nucleotide translocase (ANT) also known as ADP/ATP translocase is a protein that mechanically facilitates the exchange of ATP and ADP across the inner mitochondrial membrane. This exchange is essential for maintaining the mitochondrial membrane potential and energy metabolism. ANT has a molecular mass of approximately 30-33 kDa and is expressed in the mitochondria of virtually all eukaryotic cells where it plays an essential role in energy homeostasis.
Biological function summary

ANT functions as a transporter that is part of the mitochondrial permeability transition pore complex (mPTP). This complex plays an important role in regulating mitochondrial membrane permeability which affects processes like cell death and cellular metabolism. ANT's activity is a significant factor in regulating the mitochondrial functions since it directly influences the ATP supply to the cell.

Pathways

ANT is a core component of the oxidative phosphorylation pathway which is an important biological process for ATP production. In this pathway ANT cooperates closely with other proteins such as cytochrome c oxidase and ATP synthase to ensure efficient ATP/ADP exchange and energy production. Additionally ANT's interaction with the mPTP implicates it in the apoptotic pathway where it can influence cell survival and death decisions.

Dysfunctional ANT is linked to conditions such as ischemia-reperfusion injury and neurodegenerative diseases. In ischemia-reperfusion injury ANT's role in mPTP formation is important as excessive mPTP opening can lead to cell death. Also mutations in ANT are associated with mitochondrial myopathy and cardiomyopathy where ANT's impaired function affects cardiac and muscle energy metabolism. ANT's relationship with the voltage-dependent anion channel (VDAC) further emphasizes its role in these disease pathways.

ADP : ATP antiporter that mediates import of ADP into the mitochondrial matrix for ATP synthesis, and export of ATP out to fuel the cell (PubMed : 21586654, PubMed : 27693233). Cycles between the cytoplasmic-open state (c-state) and the matrix-open state (m-state) : operates by the alternating access mechanism with a single substrate-binding site intermittently exposed to either the cytosolic (c-state) or matrix (m-state) side of the inner mitochondrial membrane (By similarity). In addition to its ADP : ATP antiporter activity, also involved in mitochondrial uncoupling and mitochondrial permeability transition pore (mPTP) activity (PubMed : 31883789). Plays a role in mitochondrial uncoupling by acting as a proton transporter : proton transport uncouples the proton flows via the electron transport chain and ATP synthase to reduce the efficiency of ATP production and cause mitochondrial thermogenesis (By similarity). Proton transporter activity is inhibited by ADP : ATP antiporter activity, suggesting that SLC25A4/ANT1 acts as a master regulator of mitochondrial energy output by maintaining a delicate balance between ATP production (ADP : ATP antiporter activity) and thermogenesis (proton transporter activity) (By similarity). Proton transporter activity requires free fatty acids as cofactor, but does not transport it (By similarity). Also plays a key role in mPTP opening, a non-specific pore that enables free passage of the mitochondrial membranes to solutes of up to 1.5 kDa, and which contributes to cell death (PubMed : 31883789). It is however unclear if SLC25A4/ANT1 constitutes a pore-forming component of mPTP or regulates it (By similarity). Acts as a regulator of mitophagy independently of ADP : ATP antiporter activity : promotes mitophagy via interaction with TIMM44, leading to inhibit the presequence translocase TIMM23, thereby promoting stabilization of PINK1 (By similarity).
