ACSL5 also known as acyl-CoA synthetase long-chain family member 5 is an enzyme with a mass of approximately 78 kDa. This enzyme plays a critical role in lipid metabolism converting free long-chain fatty acids into fatty acyl-CoA esters ready for incorporation into complex lipids or for energy production. ACSL5 is expressed in various tissues with significant levels detected in liver adipose tissue and skeletal muscle highlighting its importance in metabolic functions.
Biological function summary

This enzyme participates in the activation of fatty acids an important step in lipid biosynthesis and degradation. ACSL5 is part of the larger acyl-CoA synthetase family known for its involvement in lipid metabolism. By converting fatty acids into their acyl-CoA derivatives it provides substrates for lipid synthesis and β-oxidation enabling efficient energy utilization and storage in cells.

Pathways

ACSL5 integrates into both the lipogenesis and β-oxidation pathways. These pathways are essential for maintaining energy balance and lipid homeostasis in cells. Catalyzes the conversion of long-chain fatty acids to their active form acyl-CoAs for both synthesis of cellular lipids, and degradation via beta-oxidation (PubMed : 17681178, PubMed : 22633490, PubMed : 24269233, PubMed : 33191500). ACSL5 may activate fatty acids from exogenous sources for the synthesis of triacylglycerol destined for intracellular storage (By similarity). Utilizes a wide range of saturated fatty acids with a preference for C16-C18 unsaturated fatty acids (By similarity). It was suggested that it may also stimulate fatty acid oxidation (By similarity). At the villus tip of the crypt-villus axis of the small intestine may sensitize epithelial cells to apoptosis specifically triggered by the death ligand TRAIL. May have a role in the survival of glioma cells.
