补充信息

Adenylate Cyclase 6 (AC6) also known as ADCY6 is an enzyme that catalyzes the conversion of ATP to cyclic AMP (cAMP) and pyrophosphate. This enzyme belongs to the class of adenylyl cyclases and has a mass of approximately 130 kDa. AC6 is widely expressed in mammalian tissues most notably in the heart brain and lungs. It plays a significant role in cellular signal transduction processes responding to G protein-coupled receptor (GPCR) activation by regulating intracellular cAMP levels.
Biological function summary

AC6 is extensively involved in cellular signaling mechanisms. It often exists as part of a larger signaling complex interacting with G proteins and other regulatory molecules. The elevation of cAMP by AC6 influences diverse physiological processes including heart rate regulation and neurotransmitter release. This establishes the importance of AC6 in both cardiac function and neural activities highlighting its impact on normal physiological states and potential pathological conditions.

Pathways

The AC6 enzyme is integral to the activation of the cAMP-dependent pathway and further influences the protein kinase A (PKA) signaling cascade. Through these pathways AC6 connects with proteins such as Gs alpha subunits and downstream elements like PKA substrates. AC6's activity helps modulate diverse cellular functions by altering cAMP concentrations which is critical for transmitting signals from GPCRs to physiological responses.

AC6 has links to heart failure and depression. Altered AC6 function can disrupt normal cardiac physiology leading to heart failure due to improper adrenergic signaling. Similarly deviations in AC6 activity have associations with depression likely through its role in neurotransmitter signaling in the brain. In these contexts AC6 interfaces with other protein targets such as beta-adrenergic receptors in the heart and serotonin receptors in the brain demonstrating its impact on disease-related pathways.

靶点信息

Catalyzes the formation of the signaling molecule cAMP downstream of G protein-coupled receptors (PubMed : 17110384, PubMed : 17916776). Functions in signaling cascades downstream of beta-adrenergic receptors in the heart and in vascular smooth muscle cells (PubMed : 17916776). Functions in signaling cascades downstream of the vasopressin receptor in the kidney and has a role in renal water reabsorption. Functions in signaling cascades downstream of PTH1R and plays a role in regulating renal phosphate excretion. Functions in signaling cascades downstream of the VIP and SCT receptors in pancreas and contributes to the regulation of pancreatic amylase and fluid secretion (By similarity). Signaling mediates cAMP-dependent activation of protein kinase PKA. This promotes increased phosphorylation of various proteins, including AKT. Plays a role in regulating cardiac sarcoplasmic reticulum Ca(2+) uptake and storage, and is required for normal heart ventricular contractibility. May contribute to normal heart function (By similarity). Mediates vasodilatation after activation of beta-adrenergic receptors by isoproterenol (PubMed : 17916776). Contributes to bone cell responses to mechanical stimuli (By similarity).
