    Inquiry: Dear Sir/ Madam, I am writing in regards to your product 'Anti-ABCB5 antibody (ab77549). I am interested in performing FACS analysis with it. I would like to ask you, however, whether the epitope recognized by the antibody is located extracellularly. Or does the antibody recognize an epitope intracellularly and thus for FACS analysis cells should be first permeabilized? With many thanks,

    I can confirm that the immunogen for this antibody is located in the internal sequence amino acids 460-471 of Human ABCB5. The amino acids are intracellular according to SwissProt:

    Therefore I suggest to use fixed and permeabilized cells. Maybe these publications can help with more information about a suitable flow cytometry protocol:
