Question

I purchased Anti-A549 antibody (ab25816) a couple of months ago and have been trying to see a response between this antibody and an A549 lysate that I have been preparing myself so far I have seen a very broad response by western blot but no response what so ever on our SPR machine. It looks like this antibody just has a VERY weak response to a lot of proteins, and is hardly specific to the A549 cell line. Has anyone else had problems with this antibody, and is there any more information you can give me?

I am sorry that you have been experiencing difficulties with this antibody in WB. In order to be able to help you further with this issue, I was hoping you could provide some additional information about your protocol and samples. Would you please provide a detailed summary of your sample preparation and WB protocol? An image would also be helpful. Once I have this information, I would be happy to offer some protocol suggestions or arrange for a solution per our Abpromise guarantee. Sorry for the delayed reply. Attached is the western blot experiment which shows that the response from the anti A549 is not at all specific and has attached itself to the majority of proteins throughout the western. This also was seen on our in-house SPR experiments where the extremely small signal is due to one the small affinity the antibody seems to have for the lysate anyway and also the fact that is sticks to the reference channel therefore giving an extremely small to no signal what so ever which is very rare for us to see. This antibody really hasn't lived up to my expectations as initially it seemed the obvious choice as an antibody against my A549 whole cell lysate.
I'm not sure what can happen here but as far as I can see it really hasn't lived up to your abcam promise.

I am sorry that you continue to experience difficulties with this antibody. The image is quite helpful, so thank you for sending that.


I am not sure what you were expecting to see from this antibody, however the datasheet outlines that the antibody will recognize more than 40 proteins by WB, including proteins from other human cell lines. The fact that it is not recognizing any protein from the E. coli lysate suggests that this antibody is functioning as expected.
