The mouse OPN kit does have a very low level cross-reactivity with human OPN. We’ve measure it to around 2-3% cross-reactivity. This is low enough I think you could measure it in your sample, but you might need to have a good control setup to ensure they are getting reliable reading.

This kit has not been optimised for use with tissue lysates but should be suitable for that purpose.

For the sample preparation, please note that there should not be more than 0.1% SDS present in the buffer.
1) Avoid using SDS or other strongly denaturing detergents. In general, non-ionic detergents, such as Triton X-100 or NP-40 are best, although zwitterionic detergents, such as CHAPS, or mild ionic detergents, such as sodium deoxycholate will work.
2) Use no more than 2% v/v total detergent
3) Avoid the use of sodium azide
4) Avoid reducing agents, such as dithiothreitol or mercaptoethanols
