ab46070 TNF-alpha rat: if the standards were ok,this indicates that the sample does not contain enough TNF alpha for detection (perhaps it has become too dilute with the washing). Do you have a good positive control sample you could try? It is possible, although notas likely, that the kit is not compatible with this sample type, I am sorry this has not been tested. For our records, I would appreciate if you are able to provide the order number and date of purchase.

ab100722 kit for IL6: I am sorry this has been tested in mouse, and not rat. I am sorry we would therefore not be able to guarantee the results from rat samples. However, the standard curve using the mouse standard provided with the kit should still be working and we can provide our guarantee for this. The protocol should be undertaken exactly as described in the protocol provided with the kit. Coud you confirm this?

I now have some further information which may help in using the MCP1 Mouse ELISA Kit (ab100722): While the optimal sample preparation procedure will need to be determined by the yourself based on knowledge of your particular samples, there are some general tips on sample preparation that may be used as a guide: The cell or tissue lysates to be used with the kit can be prepared by using conventional methods, e.g. homogenization of the cell or tissue in lysis buffer. We would recommend using the lysis buffer provided with the kit in order to perform this procedure (Cell lysate buffer). We recommend that you add some type of protease inhibitor cocktail to the lysis buffer prior to homogenization. Lysis and homogenization can be performed by glass-bead "smash", douncing, freeze thaw, sonication or crushing frozen tissue with a mortar and pestle. Following homogenization, spin down lysates to remove cell/tissue debris (5 min at 10000 x g or 10 min at 5000 g) and save the supernatant. Lysates should be frozen as soon as possible and stored at -20oC (or -80oC if possible). Spin them again before incubating with the antibody array. Determine the concentration of your lysates (for example using the bicinchoninic acid (BCA) assay) and normalize the volume of each sample used to deliver the same amount of protein for each assay. Since different cells and tissues may contain different amount of protein, as a starting point, we suggest using 500 μl of lysis buffer per 1x10^6 cells or 10 mg of tissue. You may have to adjust this based on your results. Whilst not ideal, if you would like to use an alternative lysis buffer it should meet the following specifications: have relatively low salt content (700 mM or less) does not contain sodium azide does not contain >0.1% SDS does not contain >10 mM reducing agents (beta-mercaptoethanol or dithiothreitol) no more than 2% v/v total detergent The optimal loading total protein concentration will again need to be determined by yourself but in general a good range is anywhere between 50 and 1000 μg/ml.
