The kit ab100659 TREM1 Human ELISA Kit is suitable to quantify the soluble form of human TREM-1.

Unfortunately, the concentrations used to coat the microplate and the one for the biotynilated antibody are considered to be proprietary.

The antibody’s immunogen sequence is also proprietary information. The exact epitope sequence recognized by the antibodies is unknown, we only know the antibody pair is binding somewhere on the extracellular domain (aa 21-205).

In regards to clonality, the capture antibody is monoclonal and the detection antibody is polyclonal.

The report I received from the laboratory tells us that it will be impossible to predict if this ELISA kit can detect the splice variant you are interested in, given the information we have about the antibodies.

Here is what I received from the lab:

The immunogen used to raise the capture antibody in this kit was a recombinant human TREM-1, aa 21-205 (accession # Q9NP99). The immunogen for the detection antibody was a recombinant human TREM-1, aa 1-200 (same accession #). Because epitope mapping has not been performed, the exact epitopes that the antibodies are binding to is not known. We just know that the capture antibody is binding somewhere between aa 21-205 and the detection antibody is binding somewhere between aa 1-200.

It is recommended to plot the standard curve using a log-log model as stated in the Protocol Booklet. Log-log (common log) is recommended as this model fits most data sets well. However, other graphing models can certainly be used like linear regression, 4-parameter, 2-order polynomial, etc. I would advise you to plot the data in a few models and then use whichever fits best.

I am afraid that we are not familiar with Biorad Microplate Manager 5.2 software and there are pre-settings in your machine and/or software that only the maker would know. Therefore, we unable to comment on the program's optimal settings for use with this kit. Please kindly redirect your questions to the software manufacturer.

The TREM1 Human ELISA Kit (ab100659) uses the diluent A and B in sample preparation of serum/plasma and cell culture medium/urine respectively.
The Assay Diluent A is optimized for dilution of serum and plasma samples. Assay Diluent B is for cell culture supernates, urine, and/or other sample types. The diluent separately represent the matrix of their respective sample types and should be used accordingly for optimum performance.
We are unfortunately not able to share the composition of the diluent, but the main difference is that a small amount of sodium azide is in Assay Diluent A, but not in Assay Diluent B. Sodium azide can inhibit the HRP-streptavidin function. Therefore, Assay Diluent A should not be used for dilution of the detection antibody or the HRP-streptavidin reagents.
