Interpolated concentrations of native MCP1 in rat serum plasma and cell culture supernatant samples.

The concentrations of MCP1 were measured in duplicates interpolated from the MCP1 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 7.5% plasma (citrate) 50% and spleen supernatant 2%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The MCP1 concentration was determined to be 5485 pg/mL in serum 1615 pg/mL in plasma (citrate) and 40501 pg/mL in and spleen supernatant.

How the assay works

Rat MCP1 ELISA Kit, Chemiluminescent (ab327325) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Our SimpleStep Ignite™ ELISA kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.

Rat MCP1 (ab327325) protocol summary

1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 30 min at room temperature
2. Wash
3. Add ChemiHRP development solution - incubate for 2 min
4. Read the results on a plate reader capable of luminescence detection

Acts as a ligand for C-C chemokine receptor CCR2 (By similarity). Signals through binding and activation of CCR2 and induces a strong chemotactic response and mobilization of intracellular calcium ions (By similarity). Exhibits a chemotactic activity for monocytes and basophils but not neutrophils or eosinophils (By similarity). Plays an important role in mediating peripheral nerve injury-induced neuropathic pain (By similarity). Increases NMDA-mediated synaptic transmission in both dopamine D1 and D2 receptor-containing neurons, which may be caused by MAPK/ERK-dependent phosphorylation of GRIN2B/NMDAR2B (By similarity).
