Interpolated concentrations of native IL-7 in rat serum.

The concentrations of IL-7 were measured in duplicates, interpolated from the IL-7 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-7 concentration was determined to be 251.84 pg/mL in serum.

Rat IL-7 ELISA kit (ab277704) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Rat IL-7 protein in rat serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Rat IL-7 with 1.27 pg/mL sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips

A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

IL-7 is a cytokine important for B and T cell development. In humans, IL-7 and hepatocyte growth factor form a heterodimer that functions as a pre-pro-B cell growth-stimulating factor. IL-7 is found to be a cofactor for V(D)J rearrangement of the T cell receptor beta (TCRB) during early T cell development. It can be produced locally by intestinal epithelial and epithelial goblet cells and may serve as a regulatory factor for intestinal mucosal lymphocytes. Knockout studies in mice suggested that IL-7 plays an essential role in lymphoid cell survival. Rat IL-7 shares 89% and 59% sequence homology with mouse and human respectively.

The Interleukin-7 (IL7) also known as IL-7 is a cytokine with a molecular mass of approximately 20 kDa. This protein plays an important mechanical role in cell signaling. IL7 is primarily expressed in stromal epithelial cells in the thymus and bone marrow as well as in other locations such as the liver and skin. The cytokine interacts with the IL7 receptor a heterodimer consisting of the IL7Rα chain and the common gamma chain (γc). This interaction facilitates various downstream signal transductions essential for immune cell development and homeostasis.

Biological function summary

IL7 is essential for the survival and proliferation of lymphoid cells. It acts on both T cells and B cells critical components of the adaptive immune system. The protein supports the survival of naïve and memory T cells playing a significant role in T cell homeostasis. IL7 does not operate as part of a large complex but interacts directly with its receptor to exert its function. Through its influence on T and B cell proliferation the cytokine sustains lymphoid system functionality and integrity.

Pathways

IL7 plays a pivotal role in the JAK/STAT signaling pathway which is central to cellular responses to cytokines. When IL7 binds to its receptor JAK kinases activate and promote the phosphorylation of STAT5 leading to gene transcription necessary for cell survival and proliferation. Another important pathway is the PI3K/AKT signaling cascade where the cytokine supports cell survival through anti-apoptotic signals. Through these pathways IL7 is interconnected with other cytokines such as IL2 that also function through shared or similar signaling cascades contributing to immune homeostasis.

IL7 has connections with conditions like severe combined immunodeficiency (SCID) and multiple sclerosis (MS). These involve abnormal IL7 signaling or receptor mutations that affect immune cell function. In SCID mutations in the IL7 receptor result in improper T cell development leading to severe immunodeficiency. In MS altered IL7 levels may contribute to the pathogenesis by affecting the survival and activity of autoreactive T cells. Other proteins like IL2R are also involved in these conditions as they share signaling components with IL7 and their dysregulation can lead to similar immune system disturbances.

Hematopoietic cytokine that plays an essential role in the development, expansion, and survival of naive and memory T-cells and B-cells thereby regulating the number of mature lymphocytes and maintaining lymphoid homeostasis. Mechanistically, exerts its biological effects through a receptor composed of IL7RA subunit and the cytokine receptor common subunit gamma/CSF2RG. Binding to the receptor leads to activation of various kinases including JAK1 or JAK3 depending on the cell type and subsequently propagation of signals through activation of several downstream signaling pathways including the PI3K/Akt/mTOR or the JAK-STAT5.
