      Abcam’s BNP 32 Rat in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of BNP 32 in plasma, serum, tissue extract, and cell culture supernatants.


      A BNP 32 specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently a BNP 32 specific biotinylated detection antibody is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Conjugate is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the amount of BNP 32 captured in plate.


      The entire kit may be stored at -20°C for long term storage before reconstitution - Avoid repeated freeze-thaw cycles.

    Les collaborateurs du laboratoire où les kits ELISA ab108815 et ab108816 ont été développés m'ont informé que:

    1) Les deux kits peuvent en effet être utilisés sur plasma hépariné.

    2) BNP32 et BNP45 correspondent à 2 formes de la BNP. Le croisement entre les 2 kits est d'environ 60%. D'après mes collaborateurs, il est parfois décrit que la BNP45 est plus abondante que la BNP32, et parfois il est décrit que c'est l'inverse. Le choix sur la forme de la BNP à mesurer est donc laissé à l'expérimentateur.

    Is it right that the BNP 32 Rat ELISA kit is more sensitive?

    Thank you for your enquiry.

    I can confirm that the first difference between these kits is that ab108815 detects the 32 amino acid form of BNP, andab108816 detects the larger 45 amino acid form of the protein.

    According to a literature search, the BNP prohormone is cleaved by a processing enzyme, furin, to produce mature BNP in the brain. It iscleaved by Furinata consensus sequence, Arg – X –X –Arg, and the mature BNP contains 32 amino acid residues (BNP 32). There is also the pro 45 amino acid residue form (BNP 45). BNP 45 is a major form circulating in the rat blood, and both prohormone and BNP 45 are stored in the rat atria. I am sorry I am not an expert in this field, and therefore I can suggest you may want to consider a further literature search to obtain more information.

    You are correct that the ab108815 BNP 32 kit is more sensitive, detecting down to 10 pg/ml. Whereas the ab108816 BNP 45 detects down to 30 pg/ml.
