Interpolated concentrations of native KIM-1 in mouse serum, and plasma samples.
The concentrations of KIM-1 were measured in duplicates, interpolated from the KIM-1 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 90%, plasma (citrate) 90%, plasma (heparin) 90% and plasma (EDTA) 90%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean KIM-1 concentration was determined to be 170 pg/mL in serum, 125 pg/mL in plasma (citrate), 181 pg/mL in plasma (heparin), and160 pg/mL in plasma (EDTA).

Interpolated concentrations of native mouse KIM-1 in mouse kidney supernatant, urine, and extract based on a 25%, 50%, and 50 μg/mL extract load, respectively.
The concentrations of KIM-1 were measured in duplicate and interpolated from the KIM-1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean KIM-1 concentration was determined to be 1,112 pg/mL in neat kidney supernatant, 505 pg/mL in neat urine, and 219 pg/mL in kidney extract, based on a 50 μg/mL extract load.

How the assay works
Mouse TIM 1 (KIM-1)SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Mouse TIM 1 ELISA Kit (KIM-1) ab213477 protocol summary
1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm
KIM-1, also known as Kidney Injury Molecule 1 and Hepatitis A virus cellular receptor 1 homolog (HAVcr-1), or T-cell immunoglobulin and mucin domain 1 (TIM-1) is a 283 amino acid protein that contains an N-terminal immunoglobulin-like domain and is encoded by the HAVCR1 gene. KIM-1 is a single-pass type 1 membrane protein has been identified as a receptor for hepatitis A virus. The TIM gene family participates in host immune response. Urinary KIM-1 levels are elevated in nephropathy and is thought to be a biomarker for renal damage.
