Mouse Prealbumin ELISA Kit is a single-wash 90-min SimpleStep Sandwich ELISA used to quantify Mouse Prealbumin (Transthyretin, TTR) with a sensitivity of 62.48 pg/mL. The assay uses a simple mix-wash-read protocol with just one incubation and one wash step.

Interpolated concentrations of native Prealbumin in serum, plasma (citrate), plasma (heparin) and plasma (EDTA).

The concentrations of Prealbumin were measured in duplicates, interpolated from the Prealbumin standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 25,000, plasma (citrate) 1 : 25,000, plasma (heparin) 1 : 25,000, and plasma (EDTA) 1 : 25,000. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Prealbumin concentration was determined to be 301.90 μg/mL in serum, 168.93 μg/mL in plasma (citrate), 309.51 μg/mL in plasma (heparin), and 264.54 μg/mL in plasma (EDTA).

The concentrations of Prealbumin were measured in duplicates, interpolated from the Prealbumin standard curves and corrected for sample dilution. Undiluted samples are as follows : liver supernatant 1 : 50. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Prealbumin concentration was determined to be 654.57 ng/mL in liver supernatant.

How the assay works

Mouse Prealbumin SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Assay Specificity

This kit is designed for the quantification of mouse Prealbumin. The standard protein in this kit is full length mouse Prealbumin. Native signal was detected in serum, plasma (citrate), plasma (EDTA), plasma (heparin), and tissue culture supernatant sample types. Cell extract, tissue extract, urine, saliva, CSF, and milk samples have not been tested with this kit.

1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate for 1 hr at room temperature

Prealbumin also known as transthyretin is a protein with a molecular mass of approximately 55 kDa. The liver primarily synthesizes prealbumin and it is present in the bloodstream. Functionally prealbumin transports thyroxine and retinol-binding protein-retinol complex facilitating the distribution of these molecules to target tissues. As such it plays an integral role in maintaining retinol and thyroid hormone homeostasis.
Biological function summary

Prealbumin interacts with retinol-binding protein to form a complex essential for vitamin A transportation. This complex prevents renal filtration of retinol-binding protein thereby conserving vitamin A in circulation. The protein's role extends beyond transport as it influences neurophysiological processes due to its ability to bind thyroid hormones and affect gene expression related to metabolism.
