Interpolated concentrations of native HSP60 in mouse C2C12 cell extract sample and rat C6 cell extract sample based on a 1.5 μg/mL extract load.

The concentrations of HSP60 were measured in duplicate and interpolated from the HSP60 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean HSP60 concentration was determined to be 3,456 pg/mL in C2C12 cell extract sample and 1,347 pg/mL in C6 cell extract sample.

Mouse HSP60 ELISA Kit (ab208344) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of HSP60 protein in cell culture extracts and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse HSP60 with 5.1 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips

A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

HSP60 is mitochondrial matrix protein functioning in protein folding and macromolecular assembly. HSP60 may also prevent miss-folding and promote the refolding and proper assembly of unfolded polypeptides generated under stress conditions in the mitochondrial matrix.

HSP60 also known as Heat Shock Protein 60 or Chaperonin 60 is a mitochondrial protein with a mass of about 60 kDa. It plays a critical role in protein folding and maintenance inside mitochondria. HSP60 is highly conserved across many species underlining its importance in cellular processes. Apart from mitochondria HSP60 is also present in cytosol and extracellular environments where it continues to exhibit chaperoning functions.

Biological function summary

HSP60 interacts with newly synthesized proteins preventing their aggregation and assisting in proper folding. It forms complexes with co-chaperonin proteins such as HSP10 contributing to its function. This dual complex ensures the correct assembly of mitochondrial proteins important for cellular respiration and energy production. HSP60 further assists in refolding of damaged proteins maintaining cellular proteostasis.

Pathways

HSP60 holds an important position in mitochondrial protein import and folding pathways. It acts in conjunction with other heat shock proteins such as HSP70 to facilitate translocation of proteins into mitochondria and assist in their folding once inside. HSP60 also integrates into the stress response pathway where it upregulates in response to cellular stressors aiding in the adaptation to altered physiological conditions.

HSP60 is linked to a variety of conditions including neurodegenerative disorders like Alzheimer's disease and cardiovascular diseases such as atherosclerosis. Alterations in its expression or function can lead to accumulation of misfolded proteins contributing to cell dysfunction and disease pathology. HSP60 also interacts with apoptotic proteins like BAX influencing cell survival and death pathways which may impact disease progression or resolution.

Chaperonin implicated in mitochondrial protein import and macromolecular assembly. Together with Hsp10, facilitates the correct folding of imported proteins. May also prevent misfolding and promote the refolding and proper assembly of unfolded polypeptides generated under stress conditions in the mitochondrial matrix. The functional units of these chaperonins consist of heptameric rings of the large subunit Hsp60, which function as a back-to-back double ring. In a cyclic reaction, Hsp60 ring complexes bind one unfolded substrate protein per ring, followed by the binding of ATP and association with 2 heptameric rings of the co-chaperonin Hsp10. This leads to sequestration of the substrate protein in the inner cavity of Hsp60 where, for a certain period of time, it can fold undisturbed by other cell components. Synchronous hydrolysis of ATP in all Hsp60 subunits results in the dissociation of the chaperonin rings and the release of ADP and the folded substrate protein.
