Comparison of secreted Galectin-3 in unstimulated and PMA/PHA-stimulated J774A.1 Cells.

J774A.1 cells were grown in the absence (unstimulated) or presence of Phorbol Myristate Acetate (PMA) and phytohemagglutinin (PHA) (stimulated) for 3 days. Galectin-3 was measured in 2-fold diluted cell culture supernatants of unstimulated and PMA/PHA stimulated J774A.1 and cell culture media. Measured values were interpolated from the Galectin-3 Standard Curve diluted in Sample Diluent NS and corrected for dilution factor. Mean of duplicate values +/-SD are graphed : 14 ng/mL unstimulated 38.9 ng/mL stimulated and undetectable in media.

Interpolated concentrations of Galectin-3 in mouse serum plasma (citrate) and plasma (EDTA).

The concentrations of Galectin-3 were measured in duplicate and interpolated from the Galectin-3 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Galectin-3 concentration was determined to be 11.3 ng/mL in mouse serum 13.2 ng/mL in mouse plasma (Citrate) and 12.9 ng/mL in mouse plasma (EDTA).

Mouse Galectin 3 ELISA Kit, Chemiluminescent (ab327072) is a rapid single-wash sandwich 55-min ELISA to measure Mouse Galectin 3 in Serum, EDTA Plasma, Urine, Heparin Plasma, Citrate plasma, Cell culture media with a sensitivity of 0.7 pg/mL.

How the assay works

Mouse Galectin 3 ELISA Kit, Chemiluminescent (ab327072) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Mouse Galectin 3 (ab327072) protocol summary

1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 30 min at room temperature

Galactose-specific lectin which binds IgE. May mediate with the alpha-3, beta-1 integrin the stimulation by CSPG4 of endothelial cells migration (PubMed : 15181153). Together with DMBT1, required for terminal differentiation of columnar epithelial cells during early embryogenesis. In the nucleus : acts as a pre-mRNA splicing factor. Involved in acute inflammatory responses including neutrophil activation and adhesion, chemoattraction of monocytes macrophages, opsonization of apoptotic neutrophils, and activation of mast cells. Together with TRIM16, coordinates the recognition of membrane damage with mobilization of the core autophagy regulators ATG16L1 and BECN1 in response to damaged endomembranes (By similarity). When secreted, interacts with NK cell-activating receptor NCR3/NKp30 acting as an inhibitory ligand which antagonizes NK cell attack (By similarity).
