Interpolated concentrations of native U-Plasminogen Activator in Human HEPG2 and PC-3 cell culture supernatant and urine samples.

The concentrations of U-Plasminogen Activator were measured in duplicates, interpolated from the U-Plasminogen Activator standard curves and corrected for sample dilution. Undiluted samples are as follows : HepG2 supernatant 25%, PC-3 supernatant 1 : 250, and Urine 5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean U-Plasminogen Activator concentration was determined to be 10 ng/mL in HepG2 supernatant, 865 ng/mL in PC-3 supernatant, and 73 ng/mL in Urine.

The mean U-Plasminogen Activator concentration was determined to be 439 pg/mL in serum, 732 pg/mL in plasma (citrate), 678 pg/mL in plasma (heparin), and 871 pg/ml in plasma (EDTA).

Human U-Plasminogen Activator ELISA Kit (ab226904) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of U-Plasminogen Activator protein in cell culture extracts, cell culture supernatant, cit plasma, edta plasma, hep plasma, serum, and urine. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human U-Plasminogen Activator with 4 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Urokinase also known as urokinase-type plasminogen activator (uPA) is a serine protease with a molecular mass of approximately 54 kDa. It is expressed in various tissues including the kidneys liver and blood vessels. Urokinase initially forms as pro-urokinase an inactive precursor that can convert into the active enzyme. Researchers often use assays like the direct assay to measure urokinase activity due to its role in breaking down fibrin and modulating cell adhesion and migration.
Biological function summary

Something particularly interesting about urokinase is its role in converting plasminogen to plasmin a critical enzyme in the process of fibrinolysis. Moreover it is part of a larger enzymatic complex involving the urokinase receptor (uPAR) which localizes plasminogen activation to cell surfaces. This complex mediates extracellular matrix degradation aiding in important cellular processes like tissue remodeling and wound healing.

Pathways

Urokinase plays a critical role in the fibrinolytic pathway which involves the breakdown of blood clots. It also interacts with the extracellular matrix pathway by activating plasminogen which can degrade matrix components. In these pathways urokinase closely interacts with proteins such as plasmin and its inhibitor plasminogen activator inhibitor-1 (PAI-1) which regulate the activity and localization of proteolytic events.

Urokinase is strongly linked to conditions such as deep vein thrombosis and cancer metastasis. Its overexpression or dysregulation often leads to excessive tissue degradation influencing metastasis progression. Urokinase is connected to the urokinase receptor (uPAR) in cancer facilitating cancer cell invasion and migration. Understanding its behavior in these contexts is critical for developing therapeutic strategies and targeting treatments against these diseases.
