Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Thrombin concentration was determined to be 68 μg/mL with a range of 35 – 117 μg/mL.

Interpolated concentrations of native Thrombin in human serum and plasma samples.
The concentrations of Thrombin were measured in duplicates, interpolated from the Thrombin standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 20,000, plasma (citrate) 1 : 10,000, plasma (EDTA) 1 : 10,000, and plasma (heparin) 1 : 10,000. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Thrombin concentration was determined to be 56 μg/mL in serum, 34 μg/mL in plasma (citrate), 37 μg/mL in plasma (EDTA), and 31 μg/mL in plasma (heparin).

How the assay works
Human Thrombin SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Thrombin (activated Factor II [IIa]) is a coagulation protein that has many effects in the coagulation cascade. Thrombin is a serine protease that converts soluble fibrinogen into insoluble strands of fibrin, as well as catalyzing many other coagulation-related reactions. Thrombin is in the form of alpha-thrombin that is the immediate end-product of prothrombin activation: two further thrombin products can be identified, beta- and gamma-thrombin. These are degraded forms that may arise from autodigestion of a thrombin preparation. Human Thrombin shares 81%, sequence homology with mouse and bovine as well as 80% with rat.

Biological function summary
Thrombin modulates several physiological processes beyond clot formation. It serves as a signaling molecule interacting with protease-activated receptors (PARs) to influence cell functions including proliferation migration and apoptosis. Thrombin forms part of the prothrombinase complex comprised of prothrombin activated Factor X (Xa) and Factor V on phospholipid surfaces. This complex is critical for thrombin generation during the clotting cascade. Thrombin's activity extends to involvement in wound healing and inflammation regulation.
Pathways
Thrombin significantly participates in the coagulation and fibrinolytic pathways. It not only converts fibrinogen to fibrin in the coagulation pathway but also activates inhibitors like antithrombin which regulates thrombin and other protease activities. Thrombin's interaction with fibrinolysis where tPA and plasminogen are substrates integrates clot disintegration processes. Thrombin's connectivity with proteins such as Factor VII Protein C and antithrombin outlines its diverse role in maintaining hemostatic balance.
