      Abcam’s PAI1 + tPA Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of PAI1 + tPA concentrations in plasma and serum samples.


      A PAI1 + tPA specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently a PAI1 + tPA specific biotinylated detection antibody is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Conjugate is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the amount of PAI1 + tPA captured in plate.

    I come from taiwan (asia).
    I purchased the PAI-1/tPA Human ELISA kit (ab108892) to determine the PAI-
    1/tPA complex.
    But the results are not expected, so need more help from you.
    Actually, my project is to compare the PAI-1 binding with free rtPA and
    immobilized rtPA.
    (immobilized rtAP: rtPA covalent bond with magnetic nanoparticles, it's MNP-
    rtPA)
    In vitro experiment,
    rtPA (or MNP-rtPA) incubated with PAI-1 at different molar ratio followed by
    ELISA kit was used.
    The molar ratios of rtPA/PAI-1 are 0, 0.25, 0.5, 0.75 and 1.
    As the [PAI-1] increased, i expected the PAI-1/tPA complex increased, too.
    The attatched file is the results
    The results shows that PAI-1/tPA complex was not increased with the molar
    ratio of rtPA/PAI-1 incresed
    I am also confused about the group of rtPA/PAI-1 (molar ratio: 0)
    There is no PAI-1 in this group, why the OD 450 nm was the highest?
    Please help me to resolve these quenstions or give me some commons, thanks a
    lot.

    Re Kits: ab108892- PAI-1 tPA Human ELISA kit ab46509- Thrombomodulin Human ELISA kit ab100656- Thrombopoietin Human ELISA kit I am looking for a panel of assays to detect thrombopoietin, PAI-1, and trhrombomodulin in cynomolgus monkey. I understand that the kits referenced above state their specificity to human, but can you tell me if they have been tested for cross reactivity to monkey?

    I have been in contact with the lab about the possibility of using ab108892 and ab46509 with samples from Cynomolgus Monkey. Unfortunately, the cross-reactivity is low so it will not work well for monkey samples.
