Measurement of CHIT1 activity.
Serum from ten individual healthy human female donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Osteocalcin concentration was determined to be 18.95 ng/mL with a range of 1.30 – 38.94 ng/mL.

Sandwich ELISA - Human Osteocalcin ELISA Kit (AB270202)
Interpolated concentrations of native Osteocalcin in human serum and plasma samples.
The concentrations of Osteocalcin were measured in duplicates, interpolated from the Osteocalcin standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 7.5%, plasma (citrate) 10%, plasma (heparin) 7.5%, and plasma (EDTA) 10%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Osteocalcin concentration was determined to be 34.49 ng/mL in serum, 24.94 ng/mL in plasma (citrate), 41.51 ng/ml in plasma (heparin), and 32.69 ng/ml in plasma (EDTA).

产品详情
Human Osteocalcin ELISA Kit ab270202 is a rapid single-wash 90-min Sandwich ELISA to measure Human Osteocalcin in cell culture media, citrate plasma, EDTA plasma, heparin plasma, serum. This SimpleStep sensitivity is 13.99 pg/mL.
How the assay works
Human Osteocalcin SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.
Assay Specificity
Our SimpleStep ELISA® kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.
Please refer to our protocol booklet for more details.
Human Osteocalcin ELISA Kit ab270202 protocol summary
1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm

Osteocalcin is a small secreted protein 49 amino acids produced by osteoblasts. Osteocalcin acts as a hormone impacting bone mineral absorption, glucose metabolism and insulin secretion. During bone reabsorption osteoclasts break down the bone matrix releasing bound Osteocalcin into 5 different fragments; N-terminal, Mid, C-terminal, N-terminal-Mid, and Mid-C-terminal into the serum. Circulating levels of intact Osteocalcin and Osteocalcin fragments have been found to vary significantly between healthy individuals and diseased stats such as Paget's disease, osteoporosis, fracture, and renal failure. The standard protein in this product is mature Osteocalcin.

Biological function summary
Osteocalcin plays a significant role in regulating glucose metabolism and fat deposition. It acts as a hormone from the bone and influences energy metabolism and insulin sensitivity. Osteocalcin undergoes carboxylation a post-translational modification necessary for its function in bone. Beyond its structural role demineralized osteocalcin has been found to impact pancreatic β-cell function adipose tissue and male fertility although the pathways in these processes are not entirely understood yet.
Pathways
Osteocalcin is associated with the energy metabolism and bone remodeling pathways. It interacts with the insulin signaling pathway influencing insulin secretion by pancreatic β-cells and boosting insulin sensitivity. This interaction involves proteins like insulin receptor substrate 1 (IRS1) and glucose transporter type 4 (GLUT4) which play roles in glucose uptake. Osteocalcin also connects to bone remodeling pathways by influencing osteoclast differentiation and activity through osteoprotegrin and receptor activator of nuclear factor kappa-B ligand (RANKL).
