    Lipoprotein A Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of Lipoprotein A concentrations in plasma, serum, urine, milk, CSF, cell culture supernatants, cell lysate, and tissue samples.


    A Lipoprotein A specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently a Lipoprotein A specific biotinylated detection antibody is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Conjugate is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the amount of Lipoprotein A captured in plate.


    The entire kit may be stored at -20°C for long term storage before reconstitution - Avoid repeated freeze-thaw cycles.

    Lp(a) quantification by ELISA kit ab108878 can be performed in cell lysates prepared with RIPA buffer

    HuH7 cells were incubated with exogenous Lp(a) or vehicle for 2 h, washed, pelleted, and lysed following manufacturer's protocol. Lp(a) quantification was performed in cell lysates prepared in Triton X-100 (manufacturer's recommended detergent) and, as a test, in RIPA buffer.
    Lp(a) ELISA kit ab108878 works with cell lysates prepared in RIPA buffer, yielding similar results to cell lysates prepared in Triton X-100 (figure 1).
    Furthermore, Lp(a) ELISA standard was diluted in RIPA buffer and its concentration was calculated using a standard curve (following manufacturer's protocol), yielding the expected absorbance (figure 2). This confirms RIPA buffer is compatible with Lp(a) ELISA kit ab108878.

    Dear technical team, Our customer purchased 3 of ab108878 and has some questions.   1. Her sample is plasma and following protocol page 14, she diluted her plasma sample as 1:8000. I don't know this table means dilution factor for each samples so, please let me know if it is right. 2. She used one kit and applied standard and samples but, any of well changed blue after adding substrate even standard.   She is going to try another one of kit and needs sample dilution information.   Check this please  Best regards,

    Thank you for contacting us. The information regarding the sample and standard dilutions is included under Point 6 of the Protocol “Preparation of Reagents” (Page 7). As you mention, for plasma samples, the recommended dilution is 1/8000. The table on page 14 refers to the linearity obtained for different sample types at different dilutions. The best % results for Plasma are those obtained at dilutions 1/8000 (101% of expected value). When using Serum sample, the most accurate value is obtained when diluting the samples 1/8000, for Milk the optimal dilution is 1/4000, whereas for Urine, 100% of the expected value is obtained when diluting 1/4. In the case you are still experiencing problems with any of the other kits, please do let us know, and we will be happy to help you. I hope this information is helpful to you.
