      Abcam’s Human KLK1 in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of Human KLK1 in cell culture supernatants, cell lysates, tissue homogenates, serum and plasma (heparin, EDTA).

      A KLK1 specific mouse monoclonal antibody has been precoated onto 96-well plates. Standards and test samples are added to the wells and incubated. A biotinylated detection polyclonal antibody from goat specific for KLK1 is then added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. TMB is then used to visualize the HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the Human KLK1 amount of sample captured in plate.

    Detection of human KLK1 in saliva is likely but unconfirmed by experiment. The antibodies in the assay should be capable of recognizing both isoforms 1 and 2, which are described at the UniProt entry at this link. http://www.uniprot.org/uniprot/P06870.

    This product will work for cell culture supernatants. Prepare samples as directed below: Cell culture supernates or tissue lysates: Remove particulates by centrifugation, assay immediately or aliquot and store samples at -20°C.

    Thank you for your inquiry.

    We have never tested this ELISA kit with urine samples (only serum, plasma, and tissue extracts) so we do not know if it will work. Theoretically if urine has KLK1 concentrations within the range of detection 156 pg/ml - 10000 pg/ml, this kit should work. I would suggest consulting the literature to see the known concentrations of KLK1 in urine. However, since we haven't tested this kit with urine, we would not be able to guarantee that it would work.
