Human Anti-Influenza virus A IgG ELISA Kit is an indirect ELISA for the qualitative detection of IgG class antibodies against Influenza virus A in human plasma and serum samples.
How the assay works:
A 96-well plate has been precoated with Influenza virus A antigens to ind cognate antibodies. Controls or test samples are added to the wells and incubated. Following washing, a horseradish peroxidase (HRP) labelled anti-Human IgG conjugate is added to the wells, which binds to the immobilized Influenza virus A-specific antibodies. TMB is then catalyzed by the HRP to produce a blue color product that changes to yellow after adding an acidic stop solution. The density of yellow coloration is directly proportional to the amount of Influenza virus A IgG sample captured in plate.

1. Add samples and controls to wells used. Incubate at 37°C
2. Wash each well and add prepared labeled HRP-Conjugate. Incubate at room temperature
3. After washing, add TMB substrate solution to each well. Incubate at room temperature
4. Add Stop Solution to each well. Read immediately.

Biological function summary
The influenza virus IgG antibody binds specifically to antigens present on the influenza virus A tagging them for destruction by other immune cells. It does not typically form part of larger protein complexes but operates in concert with other components of the immune system like macrophages and natural killer cells to neutralize the virus. The presence and quantity of influenza IgG antibodies are pivotal indicators of past exposure or vaccination against flu virus A in ELISA testing.
Pathways
This antibody fits into the immune system pathway where it plays a role in activating the classical complement pathway. This pathway leads to opsonization of the virus therefore improving phagocytosis efficacy by macrophages. Additionally influenza virus A IgG interacts with Fc receptors on immune effector cells facilitating processes such as antibody-dependent cellular cytotoxicity (ADCC). These interactions highlight the relationship between influenza IgG and other immune receptors in mounting an effective response.
