Comparison of IL-4 in unstimulated and PHA-M stimulated human PBMC cell supernatants.

Human PBMC cells were cultured in the absence or presence of 1.5% PHA-M for 46 hours. The concentrations of Il-4 were measured in neat supernatant samples in duplicates and interpolated from the IL-4 standard curve. The interpolated values are plotted (mean +/- SD, n=2). The mean IL-4 concentration was determined to be 174.6 pg/mL in PHA-M stimulated PBMC cell supernatant, 1.4 pg/mL in unstimulated supernatants and undetectable in media (not shown).

How the assay works

Human IL-4 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature

IL-4 is a 129 amino acids long secreted protein. IL-4 is a cytokine produced mostly by activated T lymphocytes, mast cells and basophils. It has variety of immune response modulating functions by acting on number of cell types. IL-4 participates in several B-cell activation processes. It is a co-stimulator of DNA-synthesis. It induces the expression of class II MHC molecules on resting B-cells. It enhances both secretion and cell surface expression of IgE and IgG1. It also regulates the expression of the low affinity Fc receptor for IgE (CD23) on both lymphocytes and monocytes. IL-4 positively regulates IL31RA expression in macrophages.

Cytokine secreted primarily by mast cells, T-cells, eosinophils, and basophils that plays a role in regulating antibody production, hematopoiesis and inflammation, and the development of effector T-cell responses (PubMed : 1993171, PubMed : 3016727). Induces the expression of class II MHC molecules on resting B-cells. Enhances both secretion and cell surface expression of IgE and IgG1 (PubMed : 1993171). Regulates also the expression of the low affinity Fc receptor for IgE (CD23) on both lymphocytes and monocytes (PubMed : 2521231). Positively regulates IL31RA expression in macrophages. Stimulates autophagy in dendritic cells by interfering with mTORC1 signaling and through the induction of RUFY4. In addition, plays a critical role in higher functions of the normal brain, such as memory and learning (By similarity). Upon binding to IL4, IL4R receptor dimerizes either with the common IL2R gamma chain/IL2RG to produce the type 1 signaling complex, located mainly on hematopoietic cells, or with the IL13RA1 to produce the type 2 complex, which is expressed also on nonhematopoietic cells (PubMed : 10219247, PubMed : 11526337, PubMed : 18243101). Engagement of both types of receptors initiates JAK3 and to a lower extend JAK1 phosphorylation leading to activation of the signal transducer and activator of transcription 6/STAT6 (PubMed : 7721895).
