Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Complement C2 concentration was determined to be 19.67 μg/mL with a range of 12.10 – 27.26 μg/mL.

Interpolated concentrations of native Complement C2 in human serum and plasma samples.

The concentrations of Complement C2 were measured in duplicates, interpolated from the Complement C2 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 4,000, plasma (citrate) 1 : 2,000, plasma (heparin) 1 : 1,000, and plasma (EDTA) 1 : 1,000. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Complement C2 concentration was determined to be 27.00 μg/mL in serum, 15.46 μg/mL in plasma (citrate), 11.89 μg/ml in plasma (heparin), and 12.51 μg/ml in plasma (EDTA).

The concentrations of Complement C2 were measured in duplicates, interpolated from the Complement C2 standard curves and corrected for sample dilution. Undiluted samples are as follows : HepG2 cell culture supernatant 1 : 800, milk 12.5%, and female saliva 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Complement C2 concentration was determined to be 117.47 ng/mL in HepG2 cell culture supernatant, 117.91 ng/mL in milk, and 22.87 ng/mL in female saliva.

Human Complement C2 ELISA Kit ab254501 is a rapid single-wash 90-min Sandwich ELISA to measure Human Complement C2 in cell culture supernatant, citrate plasma, EDTA plasma, heparin plasma, milk, saliva, serum. This SimpleStep sensitivity is 175.11 pg/mL.

How the assay works

Human Complement C2 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Human Complement C2 ELISA Kit ab254501 protocol summary

1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm

Complement C2 is a serine protease involved in both the classical and lectin pathways of complement activation. C2 is secreted as a zymogen. Activation of C2 by C1 leads to the proteolysis of C2 into C2a and C2b. Deficiency of C2 can lead to various types of autoimmune diseases.
