Human Cathepsin B ELISA kit (ab272205) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Cathepsin B protein in serum, plasma and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Cathepsin B with 41.8 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies

Cathepsin B is a part of a family of cysteine lysosomal proteases. Cathepsin B is synthesized as a preproenzyme in the endoplasmic reticulum and activated after transportation to the Golgi apparatus. Cathepsin B is overexpressed in invasive and metastatic cancers and is such a potential biomarker for a variety of cancers. Double knockout of cathepsin B and cathepsin L in mice is lethal due to their role in the central nervous system.

The Cathepsin B standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

The concentrations of Cathepsin B were measured in duplicates, interpolated from the Cathepsin B standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 8%, plasma (citrate) 10%, plasma (heparin) 10%, and plasma (EDTA) 10%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Cathepsin B concentration was determined to be 54.38 ng/mL in serum, 56.17 ng/mL in plasma (citrate), 55.98 ng/ml in plasma (heparin), 55.79 ng/ml in plasma (EDTA).

Interpolated concentrations of native Cathepsin B in treated human PBMC cell culture supernatant and male saliva samples.
PBMC supernatant samples were treated with 1.5% PHA-M for 46 hours. The concentrations of Cathepsin B were measured in duplicates, interpolated from the Cathepsin B standard curves and corrected for sample dilution. Undiluted samples are as follows: PBMC supernatant 4% and saliva 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Cathepsin B concentration was determined to be 103.07 ng/mL in treated PBMC supernatant and 7.00 ng/mL in male saliva.
