Interpolated concentrations of native ACE2 in human lung heart fetal intestine kidney and testes samples.

The concentrations of ACE2 were measured in duplicate and interpolated from the ACE2 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean ACE2 concentration was determined to be 26 ng/mL in lung extract 31 ng/mL in heart extract 115 ng/mL in fetal intestine 94 ng/mL in kidney and 146 ng/mL in testes extract.

The concentrations of ACE2 were measured in duplicates interpolated from the ACE2 standard curves and corrected for sample dilution. Undiluted samples are as follows : urine 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean ACE2 concentration was determined to be 75.24 ng/mL in urine.

Human ACE2 ELISA Kit, Chemiluminescent (ab327289) is a rapid single-wash sandwich 55-min ELISA to measure Human ACE2 in Serum, EDTA Plasma, Urine, Cell culture media, Tissue Extracts, Heparin Plasma, Citrate plasma with a sensitivity of 0.24 ng/mL.

How the assay works

Human ACE2 ELISA Kit, Chemiluminescent (ab327289) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Our SimpleStep Ignite™ ELISA kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.

Human ACE2 (ab327289) protocol summary

1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 30 min at room temperature
2. Wash
3. Add ChemiHRP development solution - incubate for 2 min
4. Read the results on a plate reader capable of luminescence detection

Essential counter-regulatory carboxypeptidase of the renin-angiotensin hormone system that is a critical regulator of blood volume, systemic vascular resistance, and thus cardiovascular homeostasis (PubMed : 27217402). Converts angiotensin I to angiotensin 1-9, a nine-amino acid peptide with anti-hypertrophic effects in cardiomyocytes, and angiotensin II to angiotensin 1-7, which then acts as a beneficial vasodilator and anti-proliferation agent, counterbalancing the actions of the vasoconstrictor angiotensin II (PubMed : 10924499, PubMed : 10969042, PubMed : 11815627, PubMed : 14504186, PubMed : 19021774). Also removes the C-terminal residue from three other vasoactive peptides, neurotensin, kinetensin, and des-Arg bradykinin, but is not active on bradykinin (PubMed : 10969042, PubMed : 11815627). Also cleaves other biological peptides, such as apelins (apelin-13, [Pyr1]apelin-13, apelin-17, apelin-36), casomorphins (beta-casomorphin-7, neocasomorphin) and dynorphin A with high efficiency (PubMed : 11815627, PubMed : 27217402, PubMed : 28293165). In addition, ACE2 C-terminus is homologous to collectrin and is responsible for the trafficking of the neutral amino acid transporter SL6A19 to the plasma membrane of gut epithelial cells via direct interaction, regulating its expression on the cell surface and its catalytic activity (PubMed : 18424768, PubMed : 19185582)..
