Endothelin 1 ELISA Kit ab133030 is a sandwich ELISA to measure Endothelin 1 in serum, plasma, cell culture supernatant with a sensitivity of 0.41 pg/ml.
How the assay works
Endothelin 1 specific antibody has been precoated onto 96-well plates. Standards and test samples are added to the wells and along with an HRP-conjugated I Endothelin 1 detection antibody and the microplate is then incubated at room temperature. After the removal of unbound proteins by washing, TMB is used to visualize the HRP enzymatic reaction. TMB is catalyzed by HRP to produce a colored product that changes after adding acidic stop solution. The density of coloration is directly proportional to the Endothelin 1 amount of sample captured in plate.

1. Add standard or sample to each well used. Incubate wells.
2. Aspirate and wash each well.
3. Add prepared HRP labeled secondary detector antibody. Incubate wells.
4. Aspirate and wash each well.
5. Add Chromogen Substrate Solution to each well. Immediately begin recording the color development.
Endothelins (ET) are isopeptides produced by vascular endothelium that have potent vasoconstrictive activity. The peptides are encoded by three separate genes and processed to yield 39-residue "Big ET" molecules which are further processed to the 21 amino acid sequences designated ET-1, ET-2 and ET-3. ET action is mediated via binding to two receptors, ETA and ETB. Activation of ETA increases vasoconstriction and sodium retention, thereby increasing blood pressure. Activation of ETB receptors triggers the release of vasorelaxive factors such as nitric oxide (NO) and prostanoids from endothelial cells and increases urine production and elimination, thereby lowering blood pressure.

Biological function summary
ET-1 regulates vascular tonus and blood pressure. It is part of the endothelin family that includes endothelin-2 (ET-2) and endothelin-3 (ET-3). ET-1 binds to endothelin receptors such as ETA and ETB which are G protein-coupled receptors facilitating cellular signaling processes. These receptors are abundant in smooth muscle cells and the central nervous system. The interaction of ET-1 with these receptors affects not only vascular smooth muscle contraction but also cell proliferation and production of extracellular matrix proteins.
Pathways
ET-1 plays an important role in the renin-angiotensin-aldosterone system and the nitric oxide signaling pathway. ET-1 interacts closely with angiotensin II a peptide hormone that increases blood pressure by stimulating the production of endothelin in endothelial cells. The release of nitric oxide by endothelium serves as a counter-regulatory mechanism to ET-1-mediated vasoconstriction. The balance between ET-1 and nitric oxide is vital for maintaining vascular homeostasis.
