      Canine TIMP2 ELISA Kit (ab273176) is an in-vitro enzyme-linked immunosorbent assay for the quantitative measurement of Canine TIMP2 in serum, plasma and cell culture supernatants.


      This assay employs an antibody specific for Canine TIMP2 coated on a 96-well plate. Standards and samples are pipetted into the wells and Canine TIMP2 present in a sample is bound to the wells by the immobilized antibody. The wells are washed, and biotinylated anti-Canine TIMP2 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Canine TIMP2 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

    Validation of TIMP2 ELISA kit on dog urine samples

    The kit is only validated for serum/plasma. To validate the TIMP-2 ELISA for urine samples, we spiked the TIMP-2 standard to different dilutions of urine to check recovery/matrix interference in 4 different patient groups , therefore we used a pool of healthy, CKD, AKI and Critical illness samples. We achieved all very good recoveries (70-130%). So no matrix interference was observed. We also did a serial dilution and became already nice results, only we need to repeat it for healthy and AKI with lower dilutions. But we are convinced the kit can be used for urine samples .
    In a next kit (with Abreview discount) we also want to check the urine TIMP-2 values on single patient levels to check the statistical difference between the groups and the scientific value of the biomarker in clinics as such to submit in a paper.
