    Acetylation of histones such histone H4 has been involved in the regulation of chromatin structure and the recruitment of transcription factors to gene promoters. HATs (histone acetyltransferases) and HDACs (histone deacetylases) play a critical role in controlling histone H4 actylation. Histone acetylation is tightly involved in cell cycle regulation, cell proliferation and apoptosis. Reversible acetylation of nucleosomal histone H4 is believed to correlate with potential transcriptionally activity of eukaryotic chromatin domains.

    Histone H4 Acetylation Assay Kit (ab115103) allows the user to measure global histone H4 acetylation at tremendously fast speeds and consistency, superior and safer than all other current methods. The kit is ready-to-use and provides all the essential components needed to carry out a successful assay experiment. The kit is suitable for specifically measuring global histone H4 acetylation using a variety of mammalian cells including fresh and frozen tissues, and cultured adherent and suspension cells.

Question

I'm interesting in buy some histone assay kits like: ab115103, ab115124 and others. The Kit stablish cross reactivity for mammals. I wondering if it may function with zebrafish tissues. Please let me know if I can use it with fish tissue or if you have other recomendations I will apreciate it.

Thank you for contacting us. We have not tested these kits in fish and have no testing data on this use. The kit ab115103 may be suitable for use in fish based on principle. However, the kit ab115124 may not be suitable for fish.

Question

I have a question regarding the EpiSeeker Histone H3 and H4 Acteylation Assay Kits (ab115102, ab115103). The kit instructions indicate that I will need a fluorescent microplate reader and a 60 or 100 mm plate. We do have a fluorescent microplate reader but it works only on 96 well plates. Will this be okay?

Regarding the use of a 60 or 100mm plate - this is used for disecting tissue samples if you are using them, however, once you proceed with the histone extraction, the assay is then performed in the strip wells provided with the kit. The strips can be put in the plate frame to analyze on the microplate reader.
