Developing custom rabbit monoclonal antibodies against small molecule haptens

Small molecules and polymers are common components in medicine, food, cosmetics, pesticides, narcotics and research reagents. They can also be found in the human body and in the environment.

Antibodies against this class of antigen are increasingly used in:

  • Disease diagnostics, prognosis and therapy
  • Therapeutic treatment monitoring
  • Detection and quantification of drugs and narcotics
  • Environmental monitoring

Rabbits can generate a vigorous immune response against various small molecule haptens conjugated appropriately to immunogenic carrier proteins.

Anti-Thiophosphate ester RabMAb primary antibody (ab92570) ​

Co-developed with UCSF researcher Dr. Kevan Shokat for protein kinase substrate profiling applications, this antibody provides:

  • Specific detection of thio-phosphorylated proteins
  • Context independent recognition of Ser-, Thr-, Tyr-, His-, and Asp-thiophosphorylated residues
  • Unique specificity compared to chicken antibodies. Only rabbit antibodies discriminate thiophosphate alkylation from other forms of alkylation
  • A simple, safe and environmentally friendly alternative to radioactive labeling assay

Western blot analysis using Thiophosphate Ester Specific RabMAb primary antibody (ab92570)*. In vitro kinase reaction using JNK1 protein (10ng) incubated with c-jun-GST substrate (1ug) using ATP-gamma-S followed by alkylation with PNBM (2.5mM).

Kinase reaction was carried out for 30 minutes at room temperature. PNBM alkylation (2.5mM) was for 2 hours at room temperature.

Reaction products were analyzed by western blot with thiophosphate ester specific RabMAb primary antibody (ab92570) at 1:5000, a-GST antibody confirms equal loading. Note: PNBM also reacts with cysteine residues and may decrease the affinity of other antibody-antigen interactions.

Anti-Progesterone RabMAb primary antibody (PROG-124) 

Co-developed with Pantex, a division of Bio-Analysis, the anti-Progesterone RabMAb primary demonstrates:

  • Extreme sensitivity (0.03nM) and selectivity
  • Linearity over dynamic range
  • Consistency with published test results on patient samples
