Lane 2 : C2C12 treated with 50ng/ml IFN-gamma (mouse) for 24 hours, whole cell lysate

Predicted band size: 45 kDa
Observed band size: 46 kDa why is the actual band size different from the predicted?Blocking and diluting buffer and concentration: 5% NFDM/TBST.
The expression pattern is consistent with what has been described in the literature (PMID: 30809284; PMID: 19741271).
Exposure time: 26 seconds.

Indoleamine 2,3-dioxygenase was immunoprecipitated from 0.35 mg mouse colon tissue lysate with ab277522 at 1/30 dilution (2µg in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab277522 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP)(ab131366) was used at 1/5000 dilution.
Lane 1: Mouse colon tissue lysate 10µg
Lane 2: ab277522 IP in mouse colon tissue lysate
Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab277522 in mouse colon tissue lysate
Blocking and dilution buffer and concentration: 5% NFDM/TBST.
Exposure time: 10 seconds.
