Equine adult tenocytes were fixed with 3% paraformaldehyde. Following this, the cells were permeabilized with 0.1% Triton-X-100 (1hr), blocked with 2.5% normal horse serum (20 mins) and incubated with the primary antibody for 24hrs. This antibody was tested at various concentrations (1:50, 1:100, 1:200, 1:400). Although a concentration of 1:100 was best, specific staining was seen with all concentrations.
