I performed ELISA on ovine lung tissue and plasma samples. For the tissue, the ELISA worked perfectly and the fold change trends matched what I saw in my Somascan proteomics results. For the blood, I was unable to get a reading for many samples saying that the measurement was out of range. While I got 2 blood sample readings, I did not get any blood readings for the other 22 samples. I diluted the tissue homogenate 2x fold while I did not dilute the plasma. This could be because the blood has sRAGE isoform that may differ slightly in protein configuration. Along with the cross-reactivity of human antibodies with ovine samples, this could explain why I didn't get any blood readings. The picture shows my tissue and blood ELISA measurements.
