Rat spleen gives seemingly identical positivity to that seen in other species' spleens, under same conditions (not shown).
However, the adipose tissue associated with rat spleen showed a distinct positivity in cells which are too large to be lymphocytes.
I stained a sequential section using Toluidine blue pH1(for Mast cells) and noted that the IHC-positive cells are also Tol. blue positive.
Of course, double-labelling will confirm that not only Mast cells are Granzyme-positive but that also a sub-population of Neutrophils are positive for this protein....imho.
NB: Under my conditions, the primary antibody should be further diluted.
