      Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Goat Anti-Mouse IgG Fc (HRP) (ab97265)

      IHC image of beta actin staining in normal human colon, formalin-fixed and paraffin-embedded tissue*. The section was pre-treated using pressure cooker heat mediated antigen retrieval with sodium citrate buffer (pH6) for 30mins. The section was incubated with ab8224, 3µg/ml overnight at +4°C. An HRP-conjugated secondary (Ab97265,  1/1000 dilution) was used for 1hr at room temperature. The section was counterstained with haematoxylin and mounted with DPX.

      The inset negative control image is secondary-only at 1/200 dilution.

    Inquiry: Hi Abcam, I have had great success using this secondary Ab for westerns that involve a IgG1 primary with a film exposure time of only 15s-30s. Just recently I ran a western of a recombinant protein, detecting with an IgG2a primary Ab with this as the secondary and the band took about 5 mins to develop. I know that this is not a lack of protein because I an loading a large amount of the recombinant. I also know that the primary Ab will bind this protein from earlier work. Have there been other issues reported for using this secondary with a IgG2a primary? Thanks in advance.

    This antibody should work for all IgG subclasses. Although you know your primary works, perhaps it is somewhat less efficient than the primary antibody with the IgG2a subclass?

    We have bought the following conjugates from your company. We asked the agancy that we bought about the temperature to keep them to increase the time of keeping. They told us to keep them in -20 C0 and we did for two weeks up to now, but now when we checked your website information it is recommended to be kept in 4 degree castigates.

    The datasheet of these antibodies say "Store at +4°C short term (1-2 weeks). Aliquot and store at -20°C or -80°C. Avoid repeated freeze / thaw cycles"

    This mean after receiving the antibody you can keep antibody at 4C for 1-2 weeks when using in your experiments. After you finish your experiment we recomendaliquoting the antibody 5-10 ul in vialsand storing at -20C. We suggest this to avoidthe extra freeze thawing steps, every time you use the antibody.

    If you have already stored it at -20C, this is absolutely fine!

    Thank you for contacting us.  I am still waiting for additional information regarding the strain used to generate ab106292, but I will let you know as soon as I hear back from the lab regarding this.  The other products are as follows: ab34749: 9B11 was generated against full-length recombinant Rubella capsid protein expressed in yeast S.cerevisiae. To produce the recombinant antigen, RNA was isolated from Rubella virus strain RA27/3 (GenBank: X14871.1). ab97265: This secondary antibody is specific to the Fc region of Mouse IgG and shows limited cross-reactivity with IgM and IgA. ab108750: The plates used in the kit are coated with Edmonston strain of the measles virus.  I hope this helps, please let me know if you need any additional information.
