The epitope of this rabbit anti Gli3 antibody should recognize both the full length and the cleaved isoform of Gli3 protein in the wild type lysate. Previous work has shown that both isoforms are lost in the Gli3 mutant (Xt-J) on western blot .
Abcam response
Thank you for your review of ab6050. To date, we are still unable to detect the full length protein in any lysates tested, which we believe may be due to low levels of expression in these lysates. We have previously detected the truncated isoform (80kDa) in human lung lysate and we have also detected a ~ 130kDa band similar to that seen in your Western blot, however we are unsure as to the identity of this band. We appreciate any data on this antibody.
