ab5876, a rabbit anti estrogen receptor beta has been extensively investigated on formalin-fixed and paraffin-embeded various tissue setions. Heat-induced antigen retrieval either by microwave or autoclave(citrate buffer pH6.0 or pH7.0), or proteinase K digestion, was applied to unmask antigen, and primary antibody was diluted at various concentration(from 1:100 to 1:500). However, no positive signal was detected in MCF7 xenograft, normal rat ovary, prostate and brain tissue sections. Meanwhile, estrogen receptor alpha and progesterone receptor were successfully detected in the same MCF-7 xenograft tissue section by using ab21232 and ab32085 antibodies, respectively. Frozen section was not tested yet. It is likely that this antiboy is unable to detect estrogen receptor beta in FFPE tissue section. Abcam should check the quality of ab5786 and update the necessary information to help solve this kind of troubles.
