HeLa cells were fixed for 10 minutes in PBS- 2% sucrose- 1.9% Formaldehyde and permeabilised for 10 minutes in PBS 10% sucrose-0.5% NP40. They were then blocked for 30 minutes with some PBS 10% horse serum and incubated with the primary antibody at 1:100 (diluted in blocking buffer) for an hour at RT. A control incubated with the secondary antibody only does not show any staining.
