Question

We have a customer who is experiencing troubles with your Human IgG antibody (ab7005). Please refer to below: ?We have purchased this antibody on 2 occasions now and have had no success in detecting PE fluorescence by flow cytometry in our IgG expressing cells. I note that the Immunogen is "Human IgG whole molecule" and I would like some clarification whether this means the antibody specifically recognizes the Fab portion (or hypervariable) portion of our IgG immunoglobulin alone. I also note that the specification sheet denotes under Relevance ".. absence of F(c) mediated binding" and therefore assume that this means that the antibody does not recognise the Fc portion of our secreted IgG immunoglobulin. Essentially, I am trying to detect a membrane bound IgG immunoglobulin, however I am not sure which domain (Fc or Fab) of our IgG molecule are exposed to the extracellular environment and as such whether using ab7005 is preferential to the hypervariable /Fab region only Clearly, if this is the case then we would need antibody which recognises the Fc portion in order to test our hypothesis.?. Answer

I just received this information from the source of the antibody: 1. The immunogen of Human IgG whole molecule means the antibody recognizes any epitope on the whole human IgG, not just Fab portion. 2. "..absence of F(c) mediated binding" means only F(ab')2 portion of the antibody will bind to epitopes of antigen including epitopes on the Fc portion. There is no binding to Fc receptors existing on cell surfaces. The antibody conjugated to PE is extensively cross adsorbed against serum proteins from 10 different species. Each cross that is performed removes more and more reactive antibodies from the pool until a much more narrow collection of antibody remains. While this is great for multiple label experiments, ie. to lower background against horse or goat or rabbit etc., it tends to lower the reactivity of the reagent against some specific epitopes of multiple-epitope antigen and some of the human subclass monoclonals too.
