    Beta-adrenergic receptors mediate the catecholamine-induced activation of adenylate cyclase through the action of G proteins. The beta-2-adrenergic receptor binds epinephrine with an approximately 30-fold greater affinity than it does norepinephrine.

    Palmitoylated; may reduce accessibility of Ser-345 and Ser-346 by anchoring Cys-341 to the plasma membrane. Agonist stimulation promotes depalmitoylation and further allows Ser-345 and Ser-346 phosphorylation.
    Phosphorylated by PKA and BARK upon agonist stimulation, which mediates homologous desensitization of the receptor. PKA-mediated phosphorylation seems to facilitate phosphorylation by BARK. Phosphorylated upon DNA damage, probably by ATM or ATR.
    Phosphorylation of Tyr-141 is induced by insulin and leads to supersensitization of the receptor.
    Ubiquitinated. Agonist-induced ubiquitination leads to sort internalized receptors to the lysosomes for degradation. Deubiquitination by USP20 and USP33, leads to ADRB2 recycling and resensitization after prolonged agonist stimulation. USP20 and USP33 are constitutively associated and are dissociated immediately after agonist stimulation.

Having reviewed the protocol details and done some research, it is normal to have seen a high level of the protein in your skeletal muscle samples since beta 2 Adrenergic Receptor has been previously reported to have been found in this tissue. However, I would have liked to suggest using the blocking peptide for this antibody to verify whether this antibody was indeed specific. Unfortunately, we do not have this in our catalogue. Due to this, I would suggest providing you with a free of charge replacement providing the product was purchased in the last 6 months.

I suggest ab36956 as an alternative (https://www.abcam.com/ab36956). This is also predicted to work in WB on mouse, rat and human samples. In addition, if you used this antibody and still got the same results, we do sell the blocking peptide for this antibody so you would be able to check the specificity.

Regarding ab61778, I have checked our database and have found that this antibody has been validated in many applications including Western blot and so far, we have no reason to believe that it is not specific.

In order for us to give you the best possible technical help, I would like to ask yuo a few questions.

1.Regarding your overexpressed B2AR samples, how are you doing the overexpression? How has it been validated? Are there any publications?

2. Regarding your non-overexpressed sample, what samples are you using? Are these KO cells/tissues?

The immunogen for ab61778 is a synthetic non-phosphopeptide derived from human beta 2 Adrenergic Receptor around the phosphorylation site of serine 346. Therefore based on the information available on http://www.uniprot.org/uniprot/P07550, this would indicate that the epitope is cytoplasmic.
