All lanes : Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate at 0.4 µg/ml
Predicted band size: 266 kDaWestern blot analysis of ACC1 expression using membrane enriched extracts of A549, K-562, HeLa, PC-3, HEK-293, Caco-2 and Hep G2 cell lysates. The blots were probed with ab205883 and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate. A 265 kDa band corresponding to ACC1 were observed across the cell lines tested. Along with the desired bands, non-specific bands at 120 kDa were observed across cell lines, except PC-3. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel, XCell SureLock™ Electrophoresis System and HiMark™ Pre-stained Protein Standard. Resolved proteins were then transferred onto a nitrocellulose membrane with overnight wet transfer System. The membrane was probed with the relevant primary and secondary Antibody following blocking with 5% skimmed milk. Chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate.
