No nuclear stain used, a non-primary (PBS without SLIRP antibody) control was used which showed no staining pattern. Three intensities of localisation can be seen in this image, with Type 1 being the most intense due to the greater density of mitochondria, followed by 2a then 2b (Muscle Biopsy: A Practical Approach 3rd Ed.). The uniprot page (http://www.uniprot.org/uniprot/Q9GZT3) specifies the localization is predominantly mitochondrial, as seen here. Tissue used is sub optimal with evidence of freezing artefact: focal areas lacking any staining.
