    This antibody is specific to a protein of 28 kDa. Calbindin-D-28K is also known as vitamin D-dependent calcium-binding protein or cholecalcin. Calbindin D-28K has a broad tissue distribution; however it exhibits a cell type-specific expression pattern. This antibody reacts specifically with calbindin-D-28K in brain and kidney tissues from human, bovine, goat, porcine, rabbit, dog, cat, rat, mouse, sheep, and guinea pig sources. It binds weakly to chicken calbindin-D-28K.

  • Immunohistochemical analysis of FFPE human kidney tissue, labeling Calbindin with ab82812. 

  • ab82812 staining Calbindin in Mouse brain tissue by Immunohistochemistry (PFA perfusion fixed frozen sections).

    Sections were fixed with paraformaldehyde, permeabilized with 0.2% Triton X-100 and blocked with 10% normal donkey serum + 0.3 M Glycine for 1 hour at 25°C; antigen retrieval was by heat mediation in citrate buffer. Samples were incubated with primary antibody (1/2500 in 5% serum + 0.2% Triton X-100 in PBS) for 18 hours at 4°C. An AlexaFluor® 488-conjugated Donkey anti-mouse IgG polyclonal (1/1000) was used as the secondary antibody. Counterstained with DAPI.

  • ab82812 staining rat brain (corpus callosum) sections by IHC-FoFr. The animal was perfused with 4% paraformaldehyde and further post fixed with 4% paraformaldehyde overnight. The tissues were cryoprotected with 30% sucrose and sectioned using a cryostat. Staining with ab82812 at a 1/2800 dilution in PBS-Triton (0.3%) with 0.02% azide was performed for 18h at 25°C. A donkey anti-mouse Alexa488 polyclonal antibody at 1/1000 was used as the secondary antibody.

I am looking into whether the concentration of the anti-Calbindin antibody [CB-955] (ab82812) has been determined. The antibody is provided as that collected from ascites and is therefore a mix of different proteins. The concentration of the antibody portion of this sample may therefore not have been analysed however I will try to obtain this information for you if it is available.

I, like you, have found several references where Calbindin has been used as a neuronal marker. I include a sample below. I am sorry that the referee of your paper does not support this. I would suggest doing a comprehensivejournal search to find prominent papers where anti-Calbindin antibodies have been used for this purpose and present this to the referee.

From what I can find out, ab82812 was tested with a 30 minute primary antibody incubation period at room temperature and there is no notes about whether a permeabilzation step was used or not. We do have multiply other Calbindin antibodies in our catalogue and I have looked through the Abreviews for these products and have found some information that may be helpful. Thank you for contacting Abcam. We have not tested ab82812 in free floating sections, so I am unable to send you a specific protocol for doing Free Floating section staining. However when using paraffin embedded PFA fixed slides, it is advised to use the following method: 1 - Use antigen retrieval, 10mM citrate buffer, pH6.0. 2 - Primary antibody concentrations between 1/15 -1/30 dilutions.
